赌圣退休后，赌坛再起风云，各路好手都想争夺赌王的位置。世界赌王大赛即将拉开帷幕，旨在夺冠的澳门赌霸雷泰（林国斌 饰）为了掩人耳目，想制造出另一蒙面赌圣分散众人注意力。
另一方面，自赌圣退隐后，三叔（吴孟达 饰）生活潦倒，于是前往天神村寻找赌圣的接班人。三叔发现村民阿葛 （葛民辉 饰）拥有特异功能，认为他就是赌圣的接班人，于是拉他一起闯荡江湖。在赌场内，他们遇上了泰国赌后豪姬（彭丹 饰），并得悉她正准备暗杀少女缘份（邱淑贞 饰）及她的弟弟小龙（释小龙 饰）。阿葛对缘分一见钟情，於是与三叔联手对付豪姬，救回了缘份。原来缘分是雷泰的女友，随后她带阿葛他们去见雷泰。雷泰任务阿葛正是蒙面赌圣的适合人选。赌王大赛终于开幕了！
