以现代日本为背景，描绘忍者与极道之间激烈对抗的动作漫画。1657 年，在明历大火的背后，由幕后江户守护者神赛惨蔵与始祖极道、江户第一任侠幡随院长兵卫的死斗引发的忍者与极道的宿怨，至今仍在延续。极道，既是那些被社会孤立之人的唯一归宿，同时也因犯下诸多恶行而屡遭企图消灭极道的忍者袭击。
竹本组的“幕后组长”兼“破坏之八极道”成员之一辉村极道，为了彻底消灭忍者，使用了一种能异常强化身体能力的毒品——“地狱回数券”，并在东京各处制造引人注目的骚乱，以此引诱察觉恶行而来的忍者。在这场骚乱中，一个不受法律与权力束缚、致力于阻止极道恶行的忍者团体“帝都八忍”中的一员被杀，包括“多仲忍者”在内的同伴们燃起了对极道的复仇之火。
另一方面，表面上过着高中生生活的少年“忍者（忍叶）”和表面上作为精英上班族生活的青年“极道（极美）”，因女童向动画《公主系列》的话题而意气相投，在彼此不知对方真实身份是“忍者与极道”的情况下建立了友谊。
