事业成功广受欢迎的喜剧演员乔治?西蒙斯（亚当?桑德勒 Adam Sandler 饰）发现自己身患白血病，决定趁有生之年重返单口笑话表演舞台。在一次表演中乔治结识了单口笑话表演爱好者艾拉（塞斯?罗根 Seth Rogen 饰）。穷困潦倒的艾拉一直梦想成为一名职业单口笑话演员，付不起房租的他一直借住朋友电视演员马克（詹森?舒瓦兹曼 Jason Schwartzman 饰）的客厅，同住的还有单口笑话表演爱好者利奥（乔纳?希尔 Jonah Hill 饰）。艾拉被乔治雇佣成为私人助理，说服乔治将患病消息通家人与前未婚妻劳拉（莱斯利?曼恩 Leslie Mann 饰）。乔治的白血病在新型实验药物的作用下得到控制，惊喜的他与艾拉前往旧金山表演，在城北马林县家劳拉家中二人旧情复燃。但劳拉的丈夫克拉克（埃里克?巴纳 Eric Bana 饰）此时突然返家，一切变得尴尬又糟糕……
